Part of Darley Street in Newtown was still closed at midday after police say they found a drug lab operating from within a terrace home.
A large police post has been set up on the road outside as emergency services pull apart the alleged makeshift lab.
Police and hazardous materials crews, using oxygen tanks and masks, also targeted a second home in the street.
Officers found nearly two litres of liquid, believed to be the drug GHB, and methylamphetamine tablets during the Newtown raids.
Two people, a 47-year-old man and a 37-year-old woman, were arrested at the properties.
Five raids were also rolled out on Friday morning across the Port Stephens area - including at Shoal Bay, Salt Ash and Tanilba Bay - with a large amount of ice and 6000 amphetamine tablets allegedly seized.
A shotgun and two pistols were also found.
Two women, aged 18 and 35, and a 36-year-old man have been arrested and will be questioned by police.
The raids were spurred by Operation Doboy, formed in March to investigate the supply of methylamphetamine, known as ice, in the Port Stephens area.
